11β-Hydroxysteroid-Dehydrogenase (11-DH) heißen Eukaryoten-Enzyme, die in Steroiden eine Hydroxygruppe an 11-Position zu einer Ketogruppe dehydrieren. Insbesondere 11-DH Isoform 1 katalysiert die Umwandlung von Cortisol zu Cortison und spielt eine Rolle bei der Regulation der Cortison-Aktivität. Es handelt sich um ein Membranprotein des endoplasmatischen Retikulums, das im Mensch hauptsächlich in der Leber produziert wird. Mutationen im HSD11B1-Gen sind verantwortlich für das so genannte AME-Syndrom, eine Form des Hyperandrogenismus und seltene Erbkrankheit.[2]

Katalysierte Reaktion

Cortisol + NADP+Cortison + NADPH/H+

Cortisol wird zu Cortison dehydriert. Die Typ II-Isoform verwendet Nicotinamidadenindinukleotid ( NAD+) als Kofaktor.

Einer der bekannten Hemmstoffe dieser Reaktion ist 18alpha-Glycyrrhetinsäure, die in Lakritze enthalten ist.[3][4]
